A culture method has been developed in which the medium is continuously dialysed. This method permitted the culture of human blood lymphocytes at a concentration of 20· 106/ml. Cell recovery was 60% after 6 days with a viability of 80% as judged by trypan blue exclusion. Lymphocytes cultured in this way in the presence of PPD or allogeneic cells showed a stimulation of the lactate production and the incorporation of inorganic phosphate into acid-soluble nucleotides. These changes were clearly detectable after 3–4 days and were even greater after 6 days. Sometimes the levels of glycolytic intermediates increased as well, but these effects were diminished after 6 days. The levels of ATP + ADP and the rate of oxygen consumption remained unchanged. The observed effects appeared to be completely specific: no stimulation was found when lymphocytes of Mantoux negative donors were cultured with PPD or when cells from HL-A identical siblings were cultured in a MLC.
