THE AMINOACYL-tRNA SYNTHETASES have long been identified as key enzymes in the pathway of protein synthesis in all living organisms. It is only through the mediation of these enzymes that amino acids can be activated for incorporation into growing polypeptide chains, via the formation of aminoacyl-tRNAs. Such an obvious role in providing the immediate precursors for protein synthesis does not lead us to expect additional regulatory properties of these enzymes. Nevertheless, it is clear that the synthetases do perform functions other than tRNA charging (such as the synthesis of the unusual nucleotide Ap4A for example) and a number of recent observations suggest that aminoacyl-tRNA synthetases are involved in cellular adaptation to changes in amino acid availability. In this article I aim to draw attention to such potentially important functions and to discuss whether protein phosphorylation, both of the synthetases
