Colchicine in low dosage is a weak, general anti-inflammatory agent. It is not clear at present why the drug is useful only in [human] acute gouty arthritis and a few other inflammatory disorders (such as sarcoid arthritis, and perhaps erythema nodosum with arthritis). This clinical observation may eventually be useful, however, in defining differences in the mechanism of inflammation in this group of diseases. From the effects of colchicine in various cell types, a general formulation was drawn for the action of colchicine, in which disruption of the mitotic spindle in dividing cells is a special case. Colchicine seems to interfere in cells with the organization of labile fibrillar systems (protoplasmic gels) concerned with structure and movement. Vinblastine and griseofulvin[long dash]both of which are metaphase-arresting agents[long dash]share certain other effects with colchicine that support the suggestion of a common basis for anti-mitotic and anti-inflammatory properties.
