The murine Friend leukemia virus (FLV) is being widely used to study many aspects of tumor virology. The adaptation of the leukemic tissues to in vitro growth and the demonstration that the cells had the capacity to differentiate along the erythroid pathway, provided systems which have become the standard models for studies on differentiation and erythropoiesis at the cellular, genetic, biochemical and molecular levels. The molecular pathway of the virus-induced eryth~ oleukemia, however, remains incompletely understood, notwithstanding the detailed knowledge of the genetic structures, organization and replication of retroviruses that is accumulating at a phenomenal rate. The problem stems from the fact that a number of different strains have been generated in the course of the long and varied passage history of the virus since its isolation. These strains differ from each other on the basis of their biological and biochemical properties. The question of which property (ies) is (are) necessary and sufficient to endow FLV with the capacity to produce the malignant syndrome induced by the original isolate has not been satisfactorily answered, particularly since molecularly cloned viruses differ from the wild-type virus in host range, latent period, age dependence and pathology. In this review we have tried to correlate the recent advances in virology with the cell biology and pathology in an effort to clarify the molecular basis of these differences.
