The rates of transcription of particular regions of DNA appear to be limited primarily by the frequencies with which RNA polymerase can utilize a diversity of specific DNA sequences (promoters) to initiate transcription. The 1000-fold range of RNA initiation frequencies observed in vivo is thought to reflect this diversity of promoter sequences; consequently, the investigation of transcriptional selectivity in Escherichia coli has focused largely on the relationship of promoter structure to initiation frequency. How, then, does promoter structure determine initiation frequency? A simple functional mechanism for initiation was suggested by Zillig and co-workers (Walter et al. 1967).
