We read with interest the article by Uygur et al.(2014): effects of quercetin and fish n-3 fatty acids on testicular injury induced by ethanol in rats, published recently in Andrologia. The authors found that quercetin and fish n-3 fatty acids mitigated chronic ethanol-induced testicular germ cell apoptosis and oxidative stress and enhanced antioxidant defence system. Several years ago, we reported the elevation of testicular germ cell apoptosis in chronic ethanol-treated rats (ETRs) based on TUNEL method, electron microscopy and elevated activity of caspase-3,-8 and-9 via Fas–FasL death receptor and mitochondrial signalling pathways (Eid et al., 2002; Otsuki, 2004). This enhanced apoptosis in germ cells of ETRs may be involved in testicular atrophy and infertility.However, there are some points that deserve explanation by Uygur et al. regarding the morphological observations in their article (Uygur et al., 2014). First, in fig. 1, panel c (ethanol+ quercetin group) and panel d (ethanol+ n-3 fatty acid group), the same image was shown in both panels!! How could this happen in two different testes of different animals? The same problem was repeated in fig. 2 where two images showing TUNEL-positive germ cells were the same in panels c and d of experimental groups!! These repeated images, probably accidental, should be corrected with explanation. Second, in the legend of fig. 1, the authors mentioned that S is abbreviation of Sertoli cells, while V indicates vacuole; unfortunately, the figure did not show clearly these abbreviations! Third, in the results, section of the manuscript and related to fig. 1, the authors mentioned that there was atrophy in Leydig cells of ETRs; What kind of atrophy could be observed at this low magnification? What is the meaning of Leydig cell atrophy? Uygur et al.(2014) mentioned that there is vacuolization in seminiferous tubules of ETRs testes; however, the location and nature of this vacuolization were not discussed. Therefore, it may be useful to use transmission electron microscopy for detection and analysis of morphological alterations and vacuolization in the testes of chronic ethanol-treated rats. These vacuoles may be the sites of germ cell loss after phagocytosis by Sertoli cells
