The ability to trigger a powerful, coordinated repertoire of anti-microbial reactions, including inflammation, opsonization and direct lysis, is the best known function of complement. In response, pathogens have developed an astonishing array of countermeasures to protect themselves from, and indeed to exploit, the complement system. This competition between host and parasite is investigated in the first two papers. It is now clear that the role of complement also extends well beyond this arena; there is a growing awareness of an integrated response by the body to infection and trauma. This integration is discussed in the final two papers, which describe the effect of complement components, primarily C3 and the receptors CR1 and CR2, on the development of humoral and cell-mediated responses, and the profound and unexpected influence of activated complement proteins on platelet reactivity.
