An assay of the oxidase that catalyzes the reduction of O2 to O2- was established by measuring the CL [chemiluminescence] of particulate fractions of PMN [polymorphonuclear leukocytes] in the presence of lucigenin. This CL required the addition of NAD(P)H and was very low in fractions of resting cells. In particulate fractions of PMN stimulated with phorbol myristate acetate selectively, the NADPH-dependent CL was increased. CL was linear with protein concentrations up to 100 .mu.g and was at least 10 times more sensitive for the detection of O2- than the assay based on spectrophotometric determination of O2- mediated cytochrome c reduction. CL was abolished by inactivating the enzyme at 56.degree. C.
