Random and large scale sequencing analysis of the 3'-directed cDNA library has provided a new way to study the frequency of gene expression in given tissues (1). Analysis of the 3'-directed partial messenger RNA sequences of the liver cancer cell line HepG2 that retains many liver specific markers (2) has shown that about 52% of the RNA population is redundant, representing abundantly or moderately expressed genes. Among 173 redundunt cDNAs tested, 135 were transcripts from novel genes. While studying full sizesequences of these transcripts, we noticed that c13aO7 (Accession number is Dl 1595) define the homologue gene of ribosomal protein reported in other organisms. The frequency of appearance of this clone was 6, out of 982 randomly-selected cDNA clones.The sequence that defines the cl3aO7 has been assigned to chromosome X by PCR mapping using monochromosome hybrid cell panel (Murakawa, manuscript in preparation). Northern blot of the mRNA prepared from HepG2 using the c1 3aO7 as a probe shows that the approximate nt size of this gene transcripts was 550.
