Adenosine diphosphate (ADP), alone amongst a number of nucleotides which have been tested, brings about the aggregation of blood platelets in plasma (Gaarder, Jonsen, Laland, Hellem & Owren, 1961). Other substances such as thrombin and the catecholamines also cause platelets to aggregate, but ADP is of special interest because platelets contain a relatively high concentration of its precursor, adenosine triphosphate (Born, 1958), and because there is evidence to suggest that thrombin and the catecholamines also induce aggregation through a reaction involving ADP (Kaser-Glanzmann & Liischer, 1962; Clayton & Cross, 1963). We have already investigated aggregation in plasma by a turbidimetric method (Born, 1962, a, b; Born & Cross, 1963a). To learn more about the phenomenon we have now used pigs' platelets separated from plasma and resuspended in simpler media. Some of this work has been reported briefly (Born & Cross, 1963b) and some of the results have been confirmed elsewhere (Haslam, personal communication).
