背景与目的过氧化氢(H2 O2)诱导结肠上皮细胞凋亡在炎症性肠病(IBD)的发病机制中起重要作用。成纤维细胞生长因子(FGF)15是FGF 19的人类直系同源物,据报道由结肠肌成纤维细胞分泌并增强结肠上皮的恢复,但关于FGF 19对结肠上皮细胞的功能知之甚少。本研究探讨了FGF 19对H2 O2诱导的结肠上皮细胞凋亡的抑制作用。方法采用小鼠结肠上皮细胞(YAMC),研究FGF 19对H2 O2诱导的结肠上皮细胞凋亡的保护作用。通过WST-8测定法测定细胞活力,通过Hoechst染色和裂解的caspase-3的Western印迹法测定细胞凋亡。用FGF 19预处理YAMC细胞,用H2 O2损伤YAMC细胞。结果经WST-8检测,FGF 19(50 ng/ml)预处理对H2 O2损伤的YAMC细胞有明显的保护作用。Hoechst染色显示,FGF 19可通过抑制caspase-3途径,显著降低H2 O2诱导的细胞凋亡。结论FGF 19对H2 O2诱导的YAMC细胞凋亡具有保护作用,可能与IBD的发病机制有关。FGF 19可能成为IBD治疗的一种可能策略,有待进一步研究。
Background/AimsThe apoptosis induced by hydrogen peroxide (H 2 O 2) in colonic epithelial cells is very important for the pathogenesis of inflammatory bowel disease (IBD). Fibroblast growth factor (FGF) 15, the human ortholog of FGF19, is reported to be secreted from colonic myofibroblasts and enhances colonic epithelial restitution, but little is known about the function of FGF19 to colonic epithelial cells. In the present study, we investigate the anti-apoptosis effect of FGF19 in colonic epithelial cells treated with H 2 O 2.MethodsYoung adult mouse colonic epithelial (YAMC) cells are used to investigate the protective role of FGF19. Cellular viability was determined by WST-8 assay, and apoptosis was measured by Hoechst staining and Western blotting of cleaved caspase-3. YAMC cells were pretreated by FGF19 and H 2 O 2 was used for cellular damage.ResultsWe demonstrated that pretreatment of FGF19 (50 ng/ml) significantly protects YAMC cells treated with H 2 O 2 assessed by WST-8. We also demonstrated Hoechst staining of YAMC cells and that H 2 O 2-induced apoptosis is significantly reduced by FGF19 treatment via inhibition of the caspase-3 pathway.ConclusionThese results indicate FGF19 protects YAMC cells against H 2 O 2 and might be related to the pathogenesis of IBD. Even further studies are needed–FGF19 may be one of the possible therapeutic strategies of IBD.
