A new reagent, 5,5''-dithiobis(2-nitrobenzoic acid) was synthesized and its usefulness in the determination of sulfhydryl groups demonstrated. The reagent is soluble in water at pH 8, where it reacts with sulfhydryl groups to produce an intense yellow color. The reaction was applied to blood, urine, a purified protein, and tissue extracts.
