D-raf in Droophila Signal Transduction

该项目的长期目标是确定D-raf介导的信号转导在果蝇模式的建立和完善中的作用。本项目的具体目的是：1)明确膜关联和D-raf功能在躯干信号转导通路中的作用；2)利用D-raf显性负突变确定躯干通路中D-raf活性的调控机制；3)表征D-raf功能与上游信号因子躯干、corkscrew和D-ras1之间的关系；4)表征D-raf在egf受体介导的通路中的活性。由于D-raf是Raf-1原癌基因的果蝇同源基因，因此对其发育作用的表征及其调控机制的确定将为理解高等生物的信号转导提供线索。

The long term objective of this project is to define the role of D-raf mediated signal transduction in the establishment and elaboration of pattern in Drosophila melanogaster. The specific aims of this project are as follows: 1) to define the role of membrane association and D-raf function in the Torso signal transduction pathway, 2) to define the mechanism of regulation of D-raf activity in the Torso pathway using dominant negative D-raf mutations, 3) to characterized the relationship between D-raf function and the upstream signaling factors Torso, corkscrew and D-ras1, and 4) to characterize the activity of D-raf in a EGF-receptor mediated pathway. Since D-raf is the Drosophila homologue of the Raf-1 proto-oncogene, the characterization of its developmental role and determination of its mechanism(s) of regulation will provide clues toward understanding signal transduction in higher organisms.
