Stress beeinträchtigt die Reproduktionsfähigkeit, jedoch sind die genauen Mechanismen noch weitgehend unerforscht. Neuere Erkenntnisse legen nahe, dass das zentrale Neurohormon für Stress, das Corticotropin-Releasing Hormon (CRH), über CRH Rezeptoren vom Typ I (CRH-R1) die Aktivität von GnRH (Gonadotrophin-Releasing Hormon) Neuronen inhibiert. Axone von CRH produzierenden Neuronen bilden Synapsen mit GnRH Neuronen aus. Wir möchten die Rolle des CRH-R1 für die Aktivität von GnRH Neuronen untersuchen, sowie die CRH produzierenden Nervenzellen beschreiben, die GnRH Neuronen innervieren. Zu diesem Zweck planen wir: 1) Den CRH-R1 spezifisch in GnRH Neuronen durch induzierbare Cre Rekombinase, die unter Kontrolle des GnRH Genpromoters steht, in gefloxten CRH-R1LoxP/LoxP Mäusen zu entfernen. Diese Mäuse werden dann in Bezug auf ihr Reproduktionsverhalten und die Stressbewältigung analysiert und zusätzlich psycho-physischem oder Immunstress ausgesetzt, um anschließend den Plasmaspiegel des luteinisierenden Hormons sowie die ex vivo pulsartige GnRH-Sekretion zu bestimmen. 2) Erfassung von retrograd synaptische Verbindungen auf GnRH Neuronen durch Infektion mit rekombinanten Viren (Adeno-assoziierte und Pseudotypisierte Rabies Viren). Wir erwarten eine Verringerung der durch Stress induzierten Beeinträchtigung von reproduktiven Funktionen in Mäusen, denen CRH-R1 in GnRH Neuronen fehlt, sowie die Identifizierung derjenigen Subpopulation von CRH Neuronen, die für die Stress-induzierte Beeinträchtigung verantwortlich ist.
