After the consumption of drugs of abuse or pharmaceuticals, the substances are stored permanently in dental hard tissue (enamel, dentin, carious material) and temporarily in biofilm on the surface of the teeth. The aims of project are the investigation of the incorporation of foreign substances into dental hard tissues and the analysis of non mineralised biofilm (dental plaque) and mineralised biofilm (dental calculus) to close the interval between the windows of drug detection of urine and hair. Model substances (different drugs of abuse and pharmaceuticals) will be applied to correlate the contact time and concentration on the incorporation into the different dental tissues. The influence of saliva and post-mortem changes on the tissues will also be evaluated. The uptake of drugs will be simulated in vitro with bovine dentin and enamel samples by mimicking the physiological pH-course, and in clinical studies with intra-oral sample holders. Several different dental materials (dentin, enamel, carious tissue, biofilm) of deceased shall be analysed and compared with the toxicological standard analysis in serum, urine, bile, hair, and other tissues. All dental tissues will be analysed qualitatively and quantitatively by advanced methods of liquid chromatography-tandem mass spectrometry (LC-MS/MS). High resolution Time-of-Flight mass spectrometric methods (LC-QTOF-MS) will be applied for post mortem drug screening in dental tissues.This study gathers new insights in the windows of detection of drugs in teeth to be able to conclude on a preceding drug uptake by analysis of dental tissues.
