项目摘要

当該年度は研究2年次に相当する。本年次までの間に、2005年から2014年までの間に当院で外科的切除が施行されたpT1b胃癌899例に対し、tissue microarray (TMA)を作製した。それらに対し、RhoGAPを含む融合遺伝子を検索するためのプローブ（ARHGAP6, ARHGAP10, ARHGAP26, ARHGAP42, MYO9A）を作製してFISHを施行した。その結果、58例にRhoGAP融合遺伝子が確認された。これらのRhoGAP融合遺伝子陽性胃癌の臨床的ならびに病理学的特徴を調べたところ、若年女性に多く、組織混在型の頻度が高く、静脈侵襲の頻度が低い一方で、リンパ管侵襲およびリンパ節転移の頻度が高いことがわかった。同コホートにおいて、研究者らが先行研究で確認したEBER陽性早期胃癌は、男性に多く、リンパ節転移陽性率が低いなどの特徴があったが、RhoGAP早期胃癌は、EBER陽性胃癌とは対照的な特徴を示すことが示された。さらには、印環細胞と低分化腺癌細胞が混在する組織成分と、微小腺管構造が互いに吻合を示す像からなる成分が混在する特徴的な組織像が高頻度に見いだされることが判明した。これらの研究成果をまとめて論文作成を進めている。

When the year is 2 years old, the study is equivalent. In 2005, 899 cases of pT1b gastric cancer were resected and tissue microarray (TMA) was performed. For example, RhoGAP contains a fusion gene, ARHGAP6, ARHGAP10, ARHGAP26, ARHGAP42, MYO9A. 58 RhoGAP fusion genes were identified. The clinical and pathological characteristics of RhoGAP fusion gene positive gastric cancer were adjusted according to the age, female age, high frequency of tissue mixed type, low frequency of venous invasion, high frequency of vascular invasion and node migration. In the same study, the researchers confirmed that EBER-positive early gastric cancer was more common in men, and the positive rate of node shift was lower. RhoGAP early gastric cancer was more common in men and EBER-positive gastric cancer was more common in women. In addition, it is clear that the tissue components and micro-duct structures are consistent with each other, and the tissue images with characteristics of mixing are high in frequency. The research results of this paper are published in the future.
