项目摘要

重合触媒濃度を3条件に設定して試料をラットの頭部に埋入し,埋入後1か月,2か月,4か月,9か月後にCT撮影を行い,試料埋入部を観察した.コラーゲンスポンジをφ8mm厚さ2mmにトリミングした.DVAの光重合触媒量は,カンファーキノン:第3級アミンをA群1.2:4.8w/v,B群0.6:2.4w/v,C群0.3:1.2w/vで調整し,BMP2は10μg/30μlDVAに調整した.DVA/コラーゲンスポンジ質量比は13/2とし,嫌気条件下で180秒間光照射し重合させた.8週齢のWistar系雄性ラット15匹を5匹ずつの3群に分け,麻酔下でA, B, C群の試料を頭蓋骨上正中に位置付けて骨膜と皮膚を縫合した.術後1, 2,4および9ヶ月後にX線μCT装置を用いて対象部位の観察を行った.その後,頭蓋骨ごと試料を採取し,試料の中央で前後に2等分して,前方部は脱灰後にパラフィン包埋して,ヘマトキシリン・エオジン(H-E)染色を行い,病理組織学的観察を行った.後方部はVillanueva骨染色後にMMA包埋して,非脱灰研磨標本を作製した.全組織量(TV),骨組織量(BV),PDVA量ならびに試料高さを計測し,BV/TVを算出した.μCTでは1か月後にはいずれの実験群でも骨組織の形成が見られたが,A群に比べるとB, C群の形成骨量は少なかったが,9か月後まで形態はほぼ維持された.病理組織学的には試料内に線維性組織が侵入して,既存骨表面から試料内に新生骨の部分的形成が認められた.PDVA表面には多核細胞が少数存在したが,炎症性細胞浸潤は認められなかった.BV/TVはA群ではB, C群に比べて有意に多い新生骨が見られた.また,A群の残存PDVA量はB, C群よりも有意に多く,試料高は C群よりも高かった.以上の結果から,光重合触媒量によってPDVAの吸収速度を調整可能であることが示唆された.

Total tissue volume (TV), bone volume (BV),PDVA volume and sample height were measured and BV/TV was calculated. The bone formation of group A was less than that of group B and group C after 1 month, and the morphology of group A was maintained after 9 months. Pathological histology showed that there were linear and dimensional tissue invasion in the sample, and the formation of new bone on the surface of the existing bone was recognized. There were A few multinucleated cells on the surface of PDVA, and inflammatory cell infiltration was recognized.BV/TV group A, B, C, and intentionally more new bone was observed. The residual PDVA quantity of group A is higher than that of group B and group C. As a result, it is possible to adjust the absorption rate of PDVA by adjusting the amount of photocatalyst.
