In this research, 10x genomics single cell RNA-seq was applied into the splenic NK cells and TIL(tumor infiltrating)-NK cells both from Reg-1fl/fl(control) and Reg1ΔNK(Reg conditional KO mice). We found that: 1. Deletion of Regnase-1 decreased number of NK cells. 2. Reg1ΔNK produced high amount of cytotoxic proteins, such as IFN-g;, perforin and granzymes and showed anti-tumor activity both in invo and in vitro.
