本研究を継続するにあたり、既報の洗い直しやこれまでの自身の研究成果の見直しを行った。本研究で使用予定の、マルチカラーフローサイトメトリーの導入を完了し、使用方法の確認を終了した。実際のヒト末梢血単核球細胞やヒト肝臓から抽出される肝浸潤リンパ球を使用し、実際の検体で確実なデータが得られるかの検証も施行している。大方検体採取から保存までの工程は確立されており、今後肝切除を施行予定の患者から、前向きに検体およびデータを採取していくにあたり、症例選択や実際の実働の人選を進めている。患者からの検体採取と並行して、これまで採取した検体資料を用いて標的因子の抽出を行い、同因子について細胞株を用いたin vitroならびにマウスを用いたin vivoの実験系を検討・構築している。将来的に前向きに採取した試料とデータから抽出される因子をある程度絞り込み、採取済の資料とデータで確認を進めていく。得られた実績や研究データは、こまめに学会発表や論文化していく方針。一方で、様々な背景肝から得られる肝浸潤リンパ球を用いた細胞機能解析を行い、肝細胞癌患者、転移性肝腫瘍患者、良性腫瘍患者など、患者背景に伴う肝浸潤リンパ球の細胞機能評価も実施していく。それらと免疫染色、腫瘍局在、癌部/非癌部間での違い、などを検討し、免疫抑制機構に関する新たな因子の抽出も進めていく。さらには、末梢血と肝組織間での関連性も検索し、末梢血から肝免疫を評価し得るバイオマーカーの検索も進めていく。
