本研究は新しい凝集反応によるAIDSウイルス感染の迅速血清診断法の開発である。本年度は次のような研究成果を得た。1.AIDSウイルス抗原の調製MOLT-4株にAIDSウイルスを感染させ、この感染細胞の継代培養を繰返し、この感染培養系で多量にAIDSウイルスを産生することを確めた。この培養液から蔗糖濃度勾配遠心法で分離精製し、得られたウイルスをSDS及びNP-40で処理、可溶化し、これを感作抗原とした。2.このウイルス抗原を富士粒子(ゲラチン粒子、富士レビオ社製)に感作して、これを凝集反応用抗原とした。3.以上のAIDSウイルス抗原感作ゲラチン粒子によってヒト血清中AIDSウイルス抗体との反応性をしらべた。反応は室温3時間放置後、プラスチックトレイの孔の底に生成される凝集像で判定した。その結果、米国AIDS患者血清47検体はすべて陽性であった。また対照として行った日本人健康人の血清300検体中5例が疑陽性を示した。更に日本人血友病患者85例はすべて陽性であった。IFも陽性。以上の結果は、本凝集反応は、特異性が高く、しかも簡単迅速な血清反応として、今後AIDS抗体スクリーニングに十分応用可能と考えられる。ここに実用化の基礎は殆ど確立したと考える。

This study is aimed at the development of a rapid serological method for the diagnosis of AIDS infection. This year's research results were obtained. 1. MOLT-4 strain of HIV/AIDS antigen was used to infect HIV/AIDS cells, and the culture of infected cells was recovered. The culture medium was separated and purified by sucrose concentration matching method, SDS and NP-40 treatment, solubilization, and antigen induction. 2. Antigen for anti-aggregation of anti-aggregation 3. The above AIDS antigens are sensitive to AIDS particles and anti-AIDS antibodies in serum. After the reaction is left at room temperature for 3 hours, the aggregation image is determined. The results showed that 47 serum samples of AIDS patients in China were positive. 5 of the 300 serum samples from healthy Japanese subjects were suspected positive. More than 85 Japanese hemophilia patients were positive. IF is positive. These results indicate that the specificity of this agglutination reaction is high, and the serum antibody is rapid. In the future, AIDS antibody is very useful. The foundation of this application is almost established.
