A＞レーザーマイクロダイセクション法を使用した遺伝子発現解析当該年度内に①通常の大腸がん、②通常の大腸がんの前がん病変（adenoma）、③CAC、④CACの前がん病変（dysplasia）のそれぞれのヒト臨床検体であるFFPEサンプルの確保とその症例の臨床病理学的特徴を抽出し今年度のデータと過去のデータを追加しデータベース化を進めた。腸管筋線維芽細胞（以下IMF）の代表的マーカーであるα-Smooth Muscle Actin(α-SMA)、PDGFRαの免疫染色を行い、IMFの同定の最適化を図った。レーザーマイクロダイセクションの準備と並行し他の解析方法、プロテオソーム解析、エクソソーム解析も検討している。B＞ヒト生サンプルを用いた遺伝子発現解析外科手術時に切除された大腸がん検体、通常腸管検体を用いて、酵素法による細胞を分散した細胞混濁液を作成し、フローサイトメトリーを用いて、線維芽細胞のマーカーであるPDGFRα陽性細胞のみを分離を行っている。プロトコール完成後はCAC、dysplasia症例の検体を用いてフローサイトメトリーを行う予定である。
