2022年度はAB22（CD44陽性中皮腫細胞株）を用いて、In Vitro解析を行った。CD44抗体IM7とCD44陽性細胞AB22を使用した光免疫治療（PIT）のvitroでの結果は良好であった。【抗体】CD44抗体は、抗体：IR700＝1:5の割合で混ぜ、IM7-IR700を精製。IM7/IR700比は平均2.5-3.0個程であった(IR700/IM7比に関してはLowry法にて算出)【細胞株】AB22(マウス中皮腫)：ECACCで購入　BALB/C由来 LL/2ーLUC2(マウス肺癌)：ATCCで購入　C57BL由来　今後肺での同種同所性モデルの作成を行う方針であり上記2種を購入。LL/2はvivoで肺へ移植した際のイメージングのためにLucの方を購入。【AB22】顕微鏡イメージング：4wellチャンバースライド使用. 5.0x104cells/well播種FACS(PI染色による死細胞定量)：35㎜ dish使用. 5.0x105cells/well播種　IM7は、添加後1時間程度でもイメージングできる程度にCD44抗原へ結合し、incubateの時間を6時間に延長するとさらにイメージングは良好であった。AB22の細胞膜へのCD44の発現量も多い印象であり、照射Jは45J程度で十分量であった。今後LL/2でも同様にvitroでの実験を行い、薬剤添加やNIR照射量の条件を調整する。さらに、次の段階としてマウス同種同所性モデルを作成する。
