本研究では骨感染症患者の採血検体を用いてメタボローム解析を予定している。しかしメタボローム解析は食事などの背景因子による影響を受ける可能性があり、このことは本研究の結果に大きな影響を与える可能性がある。骨感染症は、特に今回解析対象としている術後感染とすると非常に頻度が低く、その検体は貴重なものとなる。そのためメタボローム解析については現在収集している検体を用いて一回で本研究を完了させる準備をすることが重要である。この対策として、手術部位、食事、手術後の日数、既往歴、年齢、性別などのバイアスを極力除外した検体に統一することが、より精度の高い解析につながると考えられる。 当該年度においては、コントロール群も含めた採血検体をさらに収集し、より精度の高い解析に耐えうる検体を確保した。また本年度は複数の学会参加において骨感染症および、本研究にてバイオマーカーとして同定されることが予想されるTCAサイクルやスフィンゴ脂質などに関する最新の知見を得た。メタボローム解析は結果のデータ量が膨大となり、その結果の解釈をおこなうことも本研究の重要な要素であるため、知見のアップデートはその一助となることが期待される。 また上記のような適切な検体選別をおこなうため、メタボローム解析に秀でた企業・研究者とミーティングをおこない、最も再現性の高い条件を設定した。2022年度は検体収集は完了し、2023年3月にメタボローム解析を実施した。今後は解析結果の分析をおこない、学会発表・論文化をすすめていく予定である。
