同一患者より樹立した下顎歯肉扁平上皮癌患者の原発巣由来細胞株(WK2)と後発頸部リンパ節転移巣由来細胞株(WK3F)を使用して、マイクロアレイ法による解析を行った。その結果、著しく発現量の相違が認められた遺伝子(MAGEC1、MAGEA11、MACC1、HTRA1)が転移に密接に関与している可能性が示唆されたため、本研究の標的遺伝子（転移関連遺伝子候補）とした。まず、各遺伝子の強制発現細胞株、ノ ックダウン細胞株を作製し、in vitro での生物学的特性の解析、in vivo での転移実験を行うことで、標的遺伝子の転移への影響を評価することを。これらの遺伝子の影響を明らかにすることで、治療の標的遺伝子となり得る遺伝子候補を同定することが最終目標である。 2021年度と2022年度はWK2およびWK3Fの各々に対して、4種類の転移関連遺伝子候補の強制発現細胞株およびノックダウン細胞株の16種類を作製した。

The same patient was used to analyze the original cell line (WK2) and the later cell line (WK3F) of the cervical squamous cell carcinoma. The results showed that there was a correlation between the number of detected genes (MAGEC1, MAGEA11, MACC1, HTRA1) and the probability of close connection between them. The target genes of this study (candidate for shift related genes) were identified. The results showed that: (1) the stress expression cell line of each gene,(2) the production of cell line,(3) the analysis of biological characteristics in vitro,(4) the migration behavior in vivo,(5) the evaluation of the influence of migration behavior of target gene. The final goal of this study is to determine the impact of the gene on the target gene and to determine the candidate gene. Between 2021 and 2022, 16 types of stress-resistant cell lines were produced from 4 types of migration related gene candidates for WK2 and WK3F.
