内因性物質であるEstroneは薬物代謝酵素の1つであるCYP2C19により16α-hydroxyestorone(16α-HOE)へ代謝されることが報告されている。そのため、Estroneと代謝物である16α-HOEの存在比はCYP2C19の代謝活性を反映すると考えられる。本研究では内因性物質を指標として実際の薬物代謝酵素の活性と遺伝子多型の関連性を明らかにし、CYP2C19による代謝を受けるボリコナゾール(VRCZ)をモデル薬物とした体内動態の定量的予測を行うことを目的として以下の検討を実施した。<検討1>内因性物質(Estrone、16α-HOE)を指標としたCYP2C19代謝活性と遺伝子多型の関連<検討2>VRCZをモデル薬物とした内因性物質を指標とした薬物代謝酵素活性の評価〈成果〉<検討1>内因性物質(Estrone、16α-HOE)を用いたCYP2C19代謝活性と遺伝子多型の関連今回の調査で、まず5名の健常者の尿検体中のEstroneおよび16α-HOEをLC-MS/MS法を用いて定量を行った。CYP2C19による内因性物質の代謝活性と遺伝子多型の関連は明らかとされていない。その後、Estroneと16α-HOEの存在比からCYP2C19の代謝活性の評価を行った。5名の健常者での比較におい存在比の差と標準偏差から当初予定していた健常者50名(男性25名、女性25名)では不十分と判明し、健常人約100名(男性約80名、女性約20名)に変更し、募集を行うこととなった。現在際募集から10名の測定が完了している。

At present, we have completed the test of 10 international fundraisers.
