项目摘要

[目的]ペプチドホルモンであるグレリンは胃から産生され、成長ホルモン分泌促進や食欲増進など様々な作用を示す。また、漢方薬の六君子湯は、グレリンの血中濃度を上げ、食欲を増進させることが報告されている。しかし、グレリン受容体の発現亢進については下垂体と視床下部でin vitroの実験でしか報告されていない。六君子湯による末梢グレリン受容体への作用も考えられる。そこで本研究は六君子湯を与えた動物に放射性標識したグレリンを投与し、各組織へのグレリン集積を解析することで六君子湯によるグレリン受容体への作用を全身的に評価する。[方法]Fmoc固相合成法で、[Dpr^3(n-octanoyl),Lys^<11>(DOTA)]グレリン(1-10)を合成した。合成後、DOTAに放射性金属核種^<68>Gaを錯形成し放射性薬剤化した。薬剤を正常ラットと24時間絶食ラットに投与し、2時間後に各組織を取り出しガンマーカウンターで集積を比較した。またTLCを用いて血中安定性や、オートラジオグラフィーを用いてグレリン受容体に特異的な結合があるかを確認した。次に抗がん剤投与+漢方投与ラット(漢方投与群)と抗がん剤投与+水投与ラット(コントロール群)に薬剤を投与し、2時間後に各組織を取り出しガンマーカウンターで集積を比較した。またELISAで、血中グレリン濃度を調べた。[結果と考察]24時間絶食ラットでは正常のラットに比べて、脂肪の集積に有意な増加がみられ、その集積は100倍量の非放射性グレリン投与で消失したことから特異的集積と考えられる。また血中安定性を評価した結果、放射性グレリンは投与時から1.5時間後まで変化がみられなかった。膵臓を用いたオートラジオグラフィーでは膵臓に集積がみられ、その集積は非放射性グレリンによって消失したことから特異的であると考えられる。抗がん剤投与モデルラットでは、コントロール群に比べ漢方投与群での血中グレリン濃度が高く、食餌量も多かった。しかし放射性薬剤グレリンの各組織への集積には大きな違いはなかった。
