法医学において年齢推定は重要であり、歯のアミノ酸ラセミ化反応を利用した方法は、最も正確な年齢推定法の一つである。一般的に、ガスクロマトグラフィーを用いたアミノ酸光学異性体の分析には、β-シクロデキストリンカラムが用いられている。しかし、このカラムではアミノ酸光学異性体を高精度に分離する事は困難である。そこで本研究では、アミノ酸光学異性体を高精度に分離可能なChirasil-Valカラムを作製する事を目的とする。カラムの作製は、内面処理剤としてDPTMDS、Cyanopropyltriethoxysilane(CNPrTEOS)または3-Cyanopropyldimethoxydisilane(CNPrDEOS)で前処理後、スタティック法を用いてChirasil-Valを塗布した。カラムの評価は、n-Tridecane、アスパラギン酸(Asp)光学異性体標準試料及び歯を用いてガスクロマトグラフで行った。DPTMDS前処理カラムは、Chirasil-Valとの親和性が低く、液だれ現象が認められた。一方、CNPrTEOS及び3-CNPrDEOS前処理カラムは、Chirasil-Valとの高い親和性が認められた。特に、CNPrTEOS前処理したChirasil-Valカラムのn-Tridecane保持係数は2.035、理論段数は108348、段数/Mは4334であった。また、Asp光学異性体の分離度は2.30、保持時間は9分以内であった。これらの結果から、CNPrTEOS前処理により作成したChirasil-Valカラムは短時間で光化学異性体を高精度に分離可能であることが明らかとなった。今後、本研究で作製したChirasil-Valカラムを用いて歯のアミノ酸ラセミ化反応を分析し、正確な年齢推定を実施する予定である。
