当院に平成8年7月より10月までの期間に入院した肺結核患者21名、肺非定型抗酸菌症患者3名より、各患者さんの同意のもと末梢血を採取した。この末梢血よりLEUCOPREPを用いて、リンパ球分画をとりだし、RNA直接精製キット(ISOGEN日本ジーン)を用いてRNAを抽出、精製した。抽出したRNAに対しOligo dTをプライマーとしGIBCO-BRL社のReverse transcriptase(Superscript TM)を用いてcDNAを合成し、続いてpolymerase chain reaction(PCR)法にて増幅を行った。Nrampのexon2のpolymorphysmについては、患者検体24名、正常コントロール6名にて検討したが、今回の結果では欠失のないホモが100%であり、抗酸菌感染症の罹患しやすさとの相関は検討することができなかった。次にマウスにて抗酸菌に対する抵抗性に重要であることが明らかにされたtransmembrane domain2を中心にPCRを行った(PCR産物350bp)。得られたPCR産物は非変性ポリアクリルアミドゲルにて泳動を行い、その後、銀染色を行って、バンドを検出し泳動度の差が見られるかを検討した。しかし今回の検索では異常な泳動を示すバンド認められなかった。現在、The GATC1500Direct Blotting Electrophoresis Systemを用いてPCR産物を直接、Dideoxy法にて実際の塩基配列の確認を行っているが、現在3例の解析では正常の遺伝子の塩基配列と比較し相違(挿入、欠失、塩基置換等)は認められていない。

During the period from July to October of 2008, 21 tuberculosis patients and 3 pulmonary atypical acid-fast patients were admitted to the hospital. All patients agreed to take peripheral blood samples. The use of LEUCOPREP in the extraction of RNA The Reverse transcriptase(49 TM) of GIBCO-BRL was synthesized by cDNA extraction and amplified by polymerase chain reaction(PCR). Nramp exon2 polymerphysm was examined in 24 patients and 6 normal subjects. The results were 100% missing. The incidence of acid-fast bacteria infection was related to the study. The PCR product is 350bp. The PCR products were obtained from different species, such as silver stain, silver stain and silver stain. This time around, we're looking for unusual swimming patterns. Now, The GATC1500Direct Blotting Electrophoresis System was used to analyze PCR products directly and confirm the nucleotide alignment of the samples by Dideoxy method. Now, three samples were analyzed to compare the nucleotide alignment of normal samples (insertion, deletion, nucleotide substitution, etc.).
