今回の研究の目的は、S182遺伝子と脳血流量の低下の程度との関連を検討することである。S182遺伝子は、Kopans(Lancet 1995)、Tanahashiら(Lancet 1995)により、家族性アルツハイマー病患者での異常が報告され、アルツハイマー病の原因遺伝子である可能性が強く示唆されている。apoE-ε4の遺伝子が晩発性のアルツハイマー病との関連を指摘されているのに対して、S182遺伝子の場合は、若年発症の症例である。我々は、65歳以下の比較的若年発症の痴呆例15例、および痴呆の家族歴のある痴呆患者12例について、遺伝子検索を行った。S182遺伝子の既知の遺伝子異常の同定は、ScanuらがLancet(1991)に報告した方法を用いた。本研究の主旨を患者、家族に十分に説明し、同意を得た上で、対象である患者から、約2ml採血し、この血液から、PCR法でS182遺伝子のエクソン部分を増幅した。このPCR産物を、制限酵素であるBsphI、NlaIIIで処理し、得られたDNA fragmentを、agarose gel上で電気泳動した。gelをethidium bromideで染色後、紫外線下で写真撮影し、バンドを分析した。酵素処理後のDNA fragmentの切断の状態から、146Met→Leu (Kopans, Lancet 1995)、163His→Arg (TanahashiらLancet 1995)などの既知の遺伝子異常の有無を検討した。しかし、残念ながら明らかな遺伝子異常を示した症例がなかった。この補助金により、S182遺伝子の遺伝子異常の同定のための実験システムを整えることができた。本研究は、現在も継続中であり、今後遺伝子異常症例を発見し、脳血流シンチとの比較を行う予定である。痴呆

The purpose of this study is to investigate the relationship between S182 gene and low blood flow.
