実験1: 白内障患者血清中のヒト水晶体上皮蛋白に対する抗体価目的、方法;白内障患者血清中の抗水晶体蛋白抗体が何であるのか確かめるため、白内障患者より採取した水晶体上皮蛋白を抗原とし、患者血清と正常人血清とをウェスタンプロット法にて検索し、患者血清で反応性の高い抗体を調べた。結果;ヒト血清との反応は非特異性のバンドが数多く検出されるが、約70kDa及び30kDaの位置のバンドは、患者血清でより濃く染まり、正常人血清と比べ有意に検出された。結論;正常人血清に比べ上記分子量蛋白に対する抗体価が白内障患者血清で高く、白内障患者に特異的な抗体と思われた。今後の研究計画;上記の抗体に対する抗原を抽出し、実験動物に免疫し、産生された抗体が水晶体上皮細胞に毒性があるかを調べる。実験2: 水晶体器官培養下での抗体の水晶体嚢透過性目的、方法;昨年度は抗水晶体上皮蛋白抗体添加した水晶体器官培養下で、水晶体上皮細胞の障害度をトリパンブルー染色法にて検索し、間接的に抗体の嚢透過性を証明したが(国際白内障研究会(平成9年11月)および日本眼科学会総会(平成10年4月)で報告)、本年度は、同じく抗水晶体上皮蛋白抗体を添加したラット水晶体器官培養後、凍結切片を作成し、酵素抗体法を用い水晶体嚢を透過した抗体を検出し、抗体が水晶体嚢を通過することの直接的な証明を試みた。結果;現在までのところ培養後の水晶体上皮細胞に抗水晶体上皮蛋白抗体の存在が認められない。現在条件を変え、また、酵素抗体法に工夫を加え、検索中である。
