【目的】TNF-αは心不全、急性心筋梗塞等で上昇することが報告されているが、その電気生理的作用については不明な点が多い。今回TNF-αの遅延整流K電流(IK)に対する作用を検討した。【方法】単離モルモット心室筋細胞を全細胞型パッチクランプ法に供し、保持電位-40mVから、2秒のtest pulseを種々の膜電位まで加え、定常電流と末尾電位を測定した。細胞外は5.4mMK、細胞内は150mMKとし、nisoldipine(2μM)を加え、L型Ca電流を抑制した。全ての実験は37℃で行った。【結果】TNF-αは無刺激時にはIKを抑制しなかった。Isoproterenol(ISP)(20nM)にて増強時にIKを濃度依存性に抑制した(IC50 116±U/ml,Bmax107±17%)。また、抑制には電位依存性は認めなかった。さらに、Histamine(250nM)、Forskolin(500nM)で増強されたIKも同様に抑制し、あらかじめcAMP(1mM)を細胞内投与しIKを増強した場合には抑制を認めなかった。また、TNF-αの効果はphosphatase inhibitorであるsodium orthovanadate(100μM)あるいは、phosphodiestarase inhibitorであるisobutylmethylxanthine(100μM)存在下でも抑制は同様に認められた。百日咳毒素(PTX)処理後にはその効果は消失し、ceramidase inhibitorであるNOEにても、その効果は消失した。【総括】TNF-αは、1)ISP誘発性のIKsを濃度依存的に抑制した。2)Histamine、Forskolin誘発性のIKsを抑制し、cAMPで増強されたIKsは抑制せず、PTX処理にてその抑制効果は消失し、NOE処理にてもその効果は消失した。3)TNF-αが心不全、心筋梗塞時などカテコラミン分泌増加時にIKsを抑制し、活動電位時間を延長させ、催不整脈性に働く可能性があることが示唆された。

[Objective] TNF-α increased in cardiac insufficiency, acute myocardial infarction, etc., and its electrophysiological effects were unknown. This paper discusses the effect of TNF-α on delayed rectifier K current (IK). [Method] Single cell ventricular muscle cells were measured by whole-cell method with supply, hold potential of-40mV, test pulse of 2 seconds, membrane potential increase, steady current and terminal potential. Extracellular 5.4 mM K, intracellular 150 mM K, nisolidine (2μM) addition, L-type Ca current inhibition. The temperature is 37℃. [Results] TNF-α was inhibited in the absence of stimulation Isoproterenol(ISP)(20nM) inhibited IK concentration dependently (IC50 116±U/ml,Bmax107±17%). The potential dependence of inhibition is recognized. In addition, Histamine(250nM) and Forskolin(500nM) increased the inhibition of IK and cAMP(1mM) increased the intracellular administration of IK. TNF-α and phosphotransferase inhibitor were inhibited in the presence of isobutylmethylxanthine(100 μ M). Pertussis toxin (PTX) treatment is not effective, ceramidase inhibitor is not effective. [Summary] TNF-α and 1) ISP-induced IKs are concentration-dependent inhibitors. 2)Histamine and Forskolin induced IKs were inhibited, cAMP was increased, IKs were inhibited, PTX treatment inhibited IKs, NOE treatment inhibited IKs, and NOE treatment inhibited IKs. 3)TNF-α may inhibit IKs, prolong active potential time, and accelerate pulse rhythm during cardiac insufficiency and myocardial infarction.
