We deploy in vivo confocal imaging techniques in two major areas: (i) Transport processes in plants, and (ii) biosensors for small molecules and biochemical/biophysical processes. We systematically identify proteins that are responsible for the uptake and release of ions and metabolites and characterize their roles in the physiology of plants. To characterize the subcellular localization and regulation of the transport proteins, we use confocal imaging technology. We also develop and deploy genetically encoded fluorescent biosensors for ions, metabolites and signaling molecules in vivo. We pioneered the development of transporter activity sensors. To be able to record sensor output, we require highly sensitive and quantitative fluorescence imaging technologies. In my previous lab at Stanford, we used both spinning disk and selective plane illumination microscopy to study transporter regulation and biosensor responses. This application requests funds for a set of two fluorescence microscopes as well as an image data storage and processing system. After careful testing and evaluation of multiple instruments, we would like to acquire: [...] The systems will not only be used by our team, but integrated into CAi (Center for Advanced Imaging) in Düsseldorf, and thus be accessible to a wide range of scientists. Moreover we have included specific descriptions of the needs for 5 additional teams at the University and the Clinics.
