外科的に切除された大腸癌についての腫瘍組織及び正常大腸粘膜組織の採取、凍結保存、核酸及び蛋白の抽出を行った。また抽出RNAからの、cyclinD、RB、CDK4、MTS1遺伝子の発現を、quontitative PCR若しくはNothern blottingにより定量化を試みた。蛋白発現については抽出蛋白の分析に加えて、腫瘍組織のパラフィン包埋切片についてp16免疫組織染色を行い検討した。今回抽出したRNAからの遺伝子発現の検討では、MTS1遺伝子の発現強度は腫瘍と正常大腸粘膜において有意な差を認めなかった。また、cyclinD、RBについても同様であり、p16蛋白発現についても免疫染色の結果は腫瘍部と正常粘膜間に明らかな差異は認められなかった。これらの遺伝子及び蛋白発現の異常が明らかとなれば、それが"p16-CDK4/cyclinD-RB" pathwayといわれる系として捉えられる異常であるのかの検討、またそれらが臨床病理学的背景との照合にて発癌、進展に関わる因子であるのか否かの検討が期待されたが、残念ながら各実験系において有意な結果が得られなかった。現在はさらに多数の症例からの核酸、蛋白抽出と実験系の検証が急務と考えられるため鋭意これらの作業を行っている。また平行して、他臓器の癌腫についての検討も視野に入れ、過去に切除され凍結保存されている組織からの試料採取を行っているところである。
