In project A25 we will investigate the sub-compartmental nanodomain organization of the trans-Golgi network (TGN) that likely underlies the spatio-temporally controlled sorting of distinct cargos to different organelles or plasma membrane domains. Harnessing CRISPR/Cas9 knock-in cell lines for live-cell super-resolution imaging and acute protein degradation strategies, we aim to dissect the functional role of adaptor-based membrane scaffolding proteins in TGN nanodomain organization to segregate and sort distinct membrane cargoes in polarized epithelial cells.
