项目摘要

個人の身体的特徴、多様性に関する情報は、法科学検査に大変有用であると考えられる。本研究は遺伝学的要因の強いモンゴロイドに特徴的な一重瞼形成に関与する形態形成遺伝子について、マイクロサテライトを用いたゲノムワイドな相関解析を健常なヒトの二重瞼と一重瞼の群に分けて行い、眼瞼形成に寄与すると思われる遺伝子の検索を試みた。眼窩筋膜の厚薄、涙丘の有無、および写真を参照に上眼瞼溝の有無により眼瞼形態(二重瞼:43人、一重瞼:47人)が異なるDNAサンプルを、研究関与への同意を得た上で収集し、全染色体上にほぼ10-cMの間隔で存在する400種類のマイクロサテライトを用いてフラグメント解析を行った。各マイクロサテライトマーカーについて一重瞼群と二重瞼群における相関解析をx2検定にて行った。その結果1)P<0.05で相関を示すマーカーは102種類(46:一重瞼、56:二重瞼)検出された。更に、P<0.01で相関を示すマーカーは19種類(11:一重瞼、8:二重瞼)検出され、そのうちPc<0.05で有意な相関を示すものは11見られ、一重瞼と強い相関を示すマーカーは6種(D7S516,D10S547,D13S158,D15S117,D20S119,DXS1073;4.31<R.R.<19.61)、二重瞼と強い相関を示すマーカーは5種(D1S425,D2S319,D3S1580,D7S531,DXS1060;3.31<R.R<12.18)であった。これらのマーカー近傍に存在する遺伝子をNCBI Map Viewerで検索すると、一重瞼群にはD7S516,D13S158,D15S117から機能未知のhypothetical protein遺伝子が挙げられた。さらに、細胞骨格形成に関わるVimentin遺伝子などが候補として挙げられ、これらの遺伝子が機能的に眼瞼形態に関わっているか興味が示された。
