The developing tooth germs contain many kins of extracellular matrices including type I, III and IV collagens. The developmental function of these collagens, however, are not known in the tooth germs. In this project, we analyzed morphologically the function of collagen genes of tooth germ using antisense oligodeoxynucleotides (ASO) which bind complimentally to mRNA's of these collagens. The results obtained are as follows.1) In the tooth germs cultured in the medium containing either of ASO's for type I, III or IV collagen, the differentiation of tissue and the formation of dentin and enamel delayd comparing with control tooth germs cultured in BGJb medium only or that plus random sequence oligodeoxynucleotide. 2) In the tooth germs cultured in the medium containing either of ASO for type I or III collagen, the formation of dental sacs were insufficient, and the epithelial inclusion bodies in the enamel organs and the partial perforations of dental sacs were observed. 3) In the tooth germs cultured in the medium containing ASO fortype IV collagen, the formation of dentin and enamel delayd and the abnormal morphology of the cervical loops of enamel organs was observed. It is thought that these finding contribute to the understanding of developmental functions of these collagens in tooth germs.
