(1) Histological tsudy of Bursa fabricus :Bursa Fabricus basically consists of inner epithelium, lamina propria mucosa, submucosa whithin developted lymph follicles, and capsule. At 0 day of hatching, size of the follicles was measured 100mum in small one, and the follicle cortex developed slightly. At 2-3 weeks of age, the size of the follicles became lager and 220mum, the follicle cortex developed fairly. At 6 weeks of age, the size of the follicles became lager than these of 4 or 5 weeks of age and 500mum, the follicle cortex developed and wave finding between cortex and medulla was observed. At 7 weeks of age, the size of the follicles became the largest one and 700mum, the follicle cortex developed well. At 9 weeks of age, the size of the follicles became smaller one and decreased to 400mum, the follicle cortex maintained the development. At 11-12 weeks of age, the size of the follicles became 150-400mum, the decrease of the follicle cortex was observed. At 14 weeks of age, the size of the follicles became 300mum, the follicle cortex became thin.These findings suggest that the deep relationship between the growth of the quail and the development ・ decrease of the lymph follicles was recognized.(2) Immunohistochemical study :B-cells were identified in the follicle by immunohistochemistry.(3) Identification of lymphocytes :Middle Iymphocytes and mitosis were observed in the follicle cortex and small or large cells were observed in the follicle medulla by transmission electron microscopy. The cells with microvilli were identified B-cells by scanning electron microscopy.These findings suggest that the identification of the cells was possible by immunohistological methods and electron microscopy.
(1)法氏囊的组织学研究:法氏囊基本上由内上皮、粘膜固有层、粘膜下层和被膜组成,粘膜下层内有发达的淋巴滤泡。0日龄时,小卵泡大小可达100 μ m,皮质稍有发育。2-3周龄时,卵泡体积增大,220 μ m时,卵泡皮质发育良好。6周龄时,卵泡的大小大于4、5周龄和500 μ m,皮质发达,皮质与髓质间可见波形。7周龄时卵泡最大,达700 μ m,卵泡皮质发育良好。9周龄时,卵泡体积逐渐缩小,最小可达400 μ m,卵泡皮质仍保持正常发育。11-12周龄时,卵泡大小为150- 400 μ m,皮质减少。14周龄时,淋巴滤泡大小达到300 μ m,滤泡皮质变薄,说明鹌鹑的生长发育与淋巴滤泡的发育、减少有着密切的关系。(2)免疫组织化学研究:免疫组织化学法在卵泡中鉴定出B细胞。(3)淋巴细胞的鉴定:透射电镜下可见滤泡皮质内有中层淋巴细胞和有丝分裂,滤泡髓质内可见大小不等的细胞。扫描电镜观察显示,具有微绒毛的细胞为B细胞,提示免疫组化和电镜观察均能鉴别出B细胞。
