Apoptosis, or programd cell death, is the primary physiological mode of cell death. Fas antigen is a cell-surface protein that belong to the nerve growth factor/tumor necrosis factor receptor family and can mediated apoptosis. I have examined the expresion of Fas antigen on human hematopoietic cells and their progenitors. Fas antigen was detected on mature granulocytes, monocytes and erythrocytes. I found that Fas antigen were negative on majority of immature hematopoietic progenitors and is expressed along with hematopoietic differentiation. Futhermore, in the erythrocytic differentiation, Fas antigen appear at the stage of CFU-E (erythrocyute-colony forming untis) on their cell surface. Anti Fas-ontibody suprressed the CFU-E colony formation, suggesting that CFU-E have functional Fas receptor. Present data provide the notion that mature erythroid progenitors were regulated negatively by apoptosis through Fas receptor, and that hematopoietic stem cells expressed Fas antigen along with differentiation to become sensitive to regulation by Fas-mediated apoptosis.
