In vivo autoradiography of benzodiazepine receptor in congenital hydrocephalus rats

为明确先天性脑积水的发病机制，对HTX型先天性脑积水大鼠进行了苯二氮卓受体放射配体（^<125>I-Ro-16-0124; Ro）和^<123> i -碘安非他明（IMP）的体内双放射自显影。在Ro注射后60,90,120分钟采样显示，在90分钟采样最适合用于演示苯二氮卓类受体，且对血流的影响较小。苯二氮卓受体在大脑皮层高，在基底节区低。脑积水大鼠的密度较非脑积水大鼠弥漫性降低。s Ro与IMP相关性较好。大鼠在3周时进行脑脊液分流术，1周后进行同样的研究。Ro、IMP弥漫性升高，但无统计学意义。本研究提示苯二氮卓受体的体内放射自显影必须注意血流的影响，脑积水导致苯二氮卓受体随血流弥漫性降低。

To clarify the pathogenesis of congenital hydrocephalus, in vivo double autoradiography using radioligand of benzodiazepine receptor (^<125>I-Ro-16-0124 ; Ro) and ^<123>I-iodoamphetamine (IMP) was done in congenital hydrocephalus rat of HTX strain. Sampling at 60,90,120 minutes after Ro injection revealed that sampling at 90 minutes was most suitable for demonstration of benzodiazepine receptors with less infulence of blood flow. Benzodiazepine receptor was high in cerebral cortex and low in basal ganglia. Their density decreased diffusely in hydrocephalus rats than in non-hydrocephalus rat.s Ro and IMP was fairly well correlated. Cerebrospinal fluid shunting ooperation was done in rats at three weeks and same study was done one week later. It revealed diffuse increase of Ro and IMP without statistical significance. Present study revealed that in vivo autoradiography of benzodiazepine receptor must be cautious in influence of blood flow and hydorcephalus yielded diffuse decrease of benzodiazepine receptors in conjunction with blood flow.
