口蓋裂患者の場合,出生時にすでに口蓋筋の発達形成が悪いと考えられており,生後生理的な収縮が行えない場合には筋の発達がさらに阻害され,経時的に退行性変化を起こすと推察されるが充分には解明されていない。今回,我々は幼若ラットを用い,人工的に口蓋裂モデルを作成し,口蓋帆挙筋の経時的変化を形態学的・組織化学的に観察し若干の知見を得た。ウイスター系ラットを用い,口蓋裂群として,生後4週目にネンブタール腹腔内麻酔下に軟口蓋正中部を切断し,離断したまま断端を絹糸にて縫縮し口蓋裂を形成し,生後6週目,8週目,10週目に屠殺,口蓋帆挙筋を摘出した。対照群として無処理のラットの口蓋帆挙筋を生後4週目,6週目,8週目,10週目に摘出した。口蓋帆挙筋の連続凍結切片を作成し,HE,Gomoriトリクローム変法,actomyosinATPase,NADH,酸フォスファターゼ,非特異性エステラーゼの各染色を行い比較分析した。また,筋の一部をグルタルアルデヒド・カコジル酸,酸化オスミウムにて固定後,電顕用切片を作成し微細構造の変化を観察した。HE標本を画像解析装置nexus6810にて筋繊維数および筋線維断面積を測定し平均値を算出し週齢別の変化を検討した結果,各週齢の筋繊維総数の推移は対照群,口蓋裂群間に有意差を認めなかった。タイプ別筋繊維数では,口蓋裂群は対照群に較べタイプII線維数の割合が増加する傾向が認められた。筋繊維断面積では,10週目において口蓋裂群は対照群に較べ平均65.5%細小化していた。組織所見では,口蓋裂群において分葉状を呈す線維や筋原繊維の走行の乱れを認めた。口蓋裂モデルでは,非生理的条件下に置かれたことにより,筋線維の細小化のほかにタイプ比率の変化や退行性変化を生じることが示唆された。

In the case of patients with cleft flap, the development of flap tendons at birth is examined, and in the case of postnatal physiological contraction, the development of flap tendons is inhibited, and the onset of degenerative changes at birth is fully explained. In this paper, we have obtained some insights into the use of artificial tissue culture, morphological and histochemical studies of the temporal evolution of the tissue. The opening of the flap was observed at 4 weeks after birth. The soft flap was cut off at the middle part of the abdominal cavity. The soft flap was cut off at the end of the incision. The opening of the flap was observed at 6 weeks, 8 weeks and 10 weeks after birth. In contrast to the treatment, the leaves were removed at 4, 6, 8 and 10 weeks after birth. A comparative analysis of the frozen sections of the tissue was carried out by HE,Gomori, actomyosin ATPase,NADH, and acid. After fixing, a part of the tendon was prepared by electroporation, and the microstructure was observed. HE Li Wei image analysis device nexus6810 to determine the dimension of the rib and the cross-sectional area of the rib, calculate the average value of the weekly variation, and discuss the results of the weekly variation of the rib dimension, and identify the intentional difference between the crack groups. The dimension of the different tendons is increased by the tendency of the split group to increase compared with the dimension of the line II. The fracture area of the tendon was reduced by 65.5% on average after 10 weeks. The tissue was found to be divided into two groups, i.e., lobular, linear, and irregular. Under non-physiological conditions, the muscle line dimension is reduced, the ratio is changed, and the degeneration is generated.
