RAPID DETECTION OF MYCOPLASMAS BY A FILTER-IMMUNOBINDING TECHNIQUE

开发了一种过滤免疫结合（FIB）技术，其中使用了商业点板和强化硝基纤维素膜Nitro Plus 2000。当稀释支原体液体培养用作抗原时，用培养基吸附抗血清是至关重要的，以避免显著的非特异性FIB反应。吸附的抗血清对牛和禽支原体型菌株以及通过生长抑制试验鉴定的现场分离株产生特异性FIB反应。FIB法对犊牛鼻分泌物中牛分枝杆菌和牛分枝杆菌的检出率略低于常规培养法。FIB法对鸡链球菌和滑膜支原体的检出率远低于培养法，这可能是由于正常鸡口咽部支原体检出率较低。结果表明，FIB技术可用于支原体的快速诊断。

A filter-immunobinding (FIB) technique in which commercial dot-plates and enforced nitrocellulose membrane, Nitro Plus 2000, were utilized, was developed. When diluted mycoplasma liquid cultures were used as antigens, it was critical to adsorb antisera with culture medium in order to avoid significant unspecific FIB reactions. Adsorbed antisera gave rise to specific FIB reactions with bovine and avian mycoplasma Type strains as well as field isolates identified by growth inhibition test. Rate of detection of M.bovirhinis and M.bovis from nasal discharge of calves by FIB was siightly less than by the routine culture method. Detection rate of M.gallisepticum and M.synoviae by FIB much less than that obtained by culture method, probablly due to small CFU of mycoplasmas existed in the oropharynxes of normal chickens examined. It was concluded that FIB technique was useful for the rapid diagnosis of mycoplasmoses in which great number of organisms might exist.
