Synthetic Studies on Dearomatized Polycyclic Natural Products

II型聚酮生物合成途径产生大量具有部分脱芳构化结构的多环天然产物，吸引了生物化学和合成界的注意。基于新开发的与特定邻位二取代氧化腈的反应，我们开发了可行的构建特征多环骨架的方法，并应用于芳香聚酮的合成。通过这些研究，首次实现了新型五环海洋抗生素seragakinone A的全合成。

The type-II polyketide biosynthetic pathway produces a vast array of polycyclic natural products having a partially dearomatized structure, attracting the attention of the biochemical and synthetic communities. Based on the newly developed reactions with particular ortho-disubstituted nitrile oxide, we have developed viable methods for constructing the characteristic polycyclic skeleton and applied to the aromatic polyketide synthesis. Through these studies, the first total synthesis of seragakinone A, a novel pentacyclic marine antibiotic, was achieved.
