Jaw-Osteonecrosis is a serious side effect for patients with head and neck cancer who received radiation and/or chemo-therapy. It causes not only the limitation on the ability of patients to tolerate cancer therapies in some cases but also the harmful effects on the quality of life. However, current clinical strategies cannot efficiently prevent the occurrence of jaw-osteonecrosis. Therefore, effective therapy should be developed urgently. Adipose derived cells (ADC) isolated from male C3H mice were transplanted to the female mice just after the irradiation through the tail vein. Adipose derived cells (ADC) have been isolated from adipose tissue by the simple method with collagenase digestion and centrifugation. After 8 and 12 weeks of irradiation (16Gy) with cisplatin (7mg/kg), the jaw bone was harvested, and the histological and immunological analyses were performed to clarify the effects of ADC transplantation. On the other hand, the direct implantation of freshly isolated ADC with minimal amount of BMP could induce the ectopic bone formation. After 12 weeks of irradiation, theADC-transplanted mice reduced the obliteration of cell and blood-vessels in bone tissue compared with irradiated/non-transplanted mice.
