背景と目的：近年、α-Klotho蛋白質は抗老化ホルモンの一つとして注目されている。しかし、ヒトにおけるα-Klotho 蛋白質の長寿との関係やα-Klotho蛋白質の増減に関わる生活習慣については不明な点が多い。本研究は、Ｎ市の65歳健やか健診をベースとし、10年間(1996-2005年)で約3,000名が参加した「NISSIN Project」コホート研究を用いて、①血中α-Klotho 蛋白質量がその後の死亡及ぼす影響の検討と②血中α-Klotho 蛋白質量に影響を与える生活習慣の検討を行うことで、α-Klotho 蛋白質の長寿マーカーとしての意義を解明することを目的とした。方法：①死亡をケースとするケース・コーホート研究を行う②サブコーホート内で生活習慣とα-Klotho 蛋白質との関連を横断的に検討する。結果：本年度はケースとサブコーホートを抽出したデータベースの作成とα-Klotho 蛋白質の測定準備を行った。サブコーホートは、血液検体が得られる対象者2,851名しコーホート登録年度と性別で層化し、各層で対象者の20％に当たる570名をランダムサンプリングした。さらに2013年末までに209名の死亡を確認し、ケースとして抽出した。
