RNA-guided Biotin Ligases for the Analysis of Protein-Oligonucleotide Interactions

The aim of this project is the development and application of biotin ligases for the RNA-centric analysis of the interactome of specific RNAs with proteins. Subproject A) analyzes the interactions of proteins with chemically modified, therapeutically relevant antisense oligonucleotides inside the living cell. Subproject B) investigates the protein interactome of specific endogenous transcripts. For this, biotin ligases are being developed that can be specifically addressed by means of RNA conjugation, and the protein/RNA interactome is analyzed by LC-HR-MS/MS.
