Reproductive system is affected by energy regulating system. Starvation weight loss easilys amenorrhea in women. Recently, several factors which regulate appetite are discovered. We studied crosstalk of these factors toward female reproductive system. Our findings are as follows.1.Acute cerebroventricular injection of Ghrelin suppressed pulsatile LH secretion in ovariectomized rat, and this phenomenon was cancelled by co-administration of naroxon. Ghrelin suppressed GnRH srelease via beta-endorphine.2.Acute cerebroventricular injection of Orexin-A suppressed pulsatile LH secretion, and this phenomenon was cancelled by co-administration of naroxon or NPY antibody. Orexin-A suppressed GnRH srelease via beta-endorphine and NPY.Orexin-A suppressed pulsatile GnRH secretion via beta-endorphine and NPY in ovariectomized rat.3.Chronic intracerevro-ventricullar injection of Ghrelin or Orexin-A accelerated food intake and weight gain in mature female rats, however estrus cycle was not influenced … More at all. Hypothalamic orexinergic peptide such as ghrelin and orexin-A seemed not to have sufficient inhibitory action to induce anovulation. In weight loss and amenorrheic situation, many signals which control energy homeostasis including hypothalamic orexinergic peptides and peripheral originated circulating leptin would be integrated and work together to stop ovulation.4.Leptin stimulated gonadotropin secretion in juvenile rat pituitary cells, and suitable leptin concentration for this stimulatory effect shifed in accordance with development ; Juvenile rat pituitary was more sensitive to leptin than latter ages. Messenger RNA of functional leptin receptor(OB-Rb) was highly expressed at juvenile age compared with latter ages in female rats pituitary. On the other hand, serum leptin concentration increased though pubertal development in contrast to OB-Rb mRNA expression in the pituitary. In conclusion, juvenile female rat pituitary might be sensitive to the increase of serum leptin concentration preparing puberty. Less
