本研究は、アトピー性皮膚炎におけるコラーゲントリペプチドの効果を検討した。申請者はこれまで、乾燥肌モデルマウスにおいてコラーゲントリペプチドが真皮のヒアルロン酸のヒアルロン酸産生を促し、乾燥にともなう痒みを改善することを明らかにした。その機序を明らかにするために本研究では、炎症性サイトカインであるIL-13、TNF-α、IFN-γを添加したヒト表皮角化細胞にコラーゲントリペプチドを添加し、TARC、TSLPの産生を測定することにより、コラーゲントリペプチドが炎症時の表皮角化細胞に与える影響について解析を行った。TARCの産生をRT-PCRで測定した結果、コラーゲントリペプチドを添加していない表皮角化細胞と比べて、TARCの産生が抑制されていた。また、TSLPについても同様の結果が得られた。ELISA法でTARCの蛋白定量を行った結果、コラーゲントリペプチドにより表皮角化細胞のTARCが低下する傾向がみられたが、有意差はみられなかった。同様に、TSLPをウエスタンブロット法で確認したところ、コラーゲントリペプチドの添加によりTSLPのタンパク量が減少することが明らかになった。さらに、13人のアトピー性皮膚炎患者に対して、コラーゲントリペプチド(7人）、コラーゲンペプチド(6人）を12週間投与し、SCORAD、角質水分量、痒みの評価、血清中のTARC、IgE、LDHおよび好酸球数を測定した。その結果、コラーゲントリペプチド投与群でのみ、内服投与開始前と比べて皮疹面積、SCORAD、TEWLが改善していた。コラーゲントリペプチド群では投与前と比べてTARCの減少がみられた。血清中のIgE、LDHおよび好酸球数についてはいずれの群でも投与前後で有意差は認めなかった。
