本研究では,まず,35gに調整されたmodified aneurysm clipを用いてラット脊髄損傷モデルを作製し,後肢の運動機能および機械刺激による逃避閾値 (withdrawal threshold; WT)の経時的評価を行った.後肢運動機能は,Basso Beattie Bresnahan (BBB ) locomotor rating scaleを用いて,後肢WTは,電子痛覚測定装置 (Electrovonfrey, model 2290, IITC Inc, Woodland Hills, CA)を用いて評価した.その結果,後肢運動機能については,モデル作製直後よりBBBスコアの著明な低下を認め,4週間の追跡期間中,低下した状態が維持された.次に,後肢WTについては,モデル作製後2週間目より,著明な低下が確認され,その後,2週間同様の状態が確認された.次に,ラット用脊髄硬膜外刺激電極を入手することが困難であったため,同電極の開発を行った.2極の円盤型プラチナ電極が配置された試作品①(幅2.0mm,長さ6mm,厚さ1.0mm)を2匹のラットに対し留置したが,いずれのラットにおいても,直後より,後肢運動機能の著明な低下を認め,電極留置に伴い脊髄損傷を生じたものと判断した.その後,幅1.8mm,長さ6mm,厚さ0.7mmに改良した試作品②を作製し,同様に3匹のラットに対し留置したが,やはり脊髄損傷による下肢運動機能の低下を認めた.これ以上細径の電極の作成は困難であったことから,ラットのサイズを300g程度と大きくして,再度,電極を留置したが,脊髄損傷を生じてしまった.このように,適切なラット用脊髄硬膜外刺激電極を作製することが困難であったことから,本研究の主目的である,ラット脊髄損傷モデルを用いた脊髄刺激の有効性評価を行うことができなかった.
