マウスマクロファージ様細胞株、P388【D_1】、のあるクローンはその培養液中に、IL-1とともにIL-1抑制因子をも産生している事が認められた。このクローン株由来のIL-1抑制因子はヒト,マウスいずれの由来のIL-1の作用に対しても抑制活性を示した。この抑制因子はIL-1の作用のみに特異的でIL-2や、他のサイトカインの作用に対しては抑制活性を示さなかった。またIL-1の産生にも影響を与えなかった。ゲル濾過により、このIL-1抑制因子の分子量は約50KDと推定されたが、DEAEセルロースによる分離法においては、Nacl濃度各々、170mM,350mMに溶出される2相性のピークを示した。170mMに溶出される抑制活性はIL-1の作用のみならず、IL-2のそれをも抑制したが、350mMへの溶出画分はIL-1の作用のみを特異的に抑制した。またクロマトフォーカシングにより、170mM,350mM溶出画分の抑制因子の等電点はそれぞれ、59、52〜53であった。さらに170mM溶出画分の抑制因子はPH3.0の酸処理もしくは56℃、60分間の加熱処理によりその活性を失うが、350mM溶出画分は同様の処理に対し安定であった。

The production of IL-1 and IL-1 inhibitory factors in culture medium of P388 [D_1] cell line The inhibitory activity of IL-1 in these strains is demonstrated by the action of IL-1 in these strains. These inhibitory factors are specific for IL-1 and IL-2, and their inhibitory activity is also demonstrated by their inhibitory effects on IL-1 and IL-2. IL-1 and its effects The molecular weight of IL-1 inhibitor was estimated to be about 50KD by filtration and DEAE separation. The concentration of Nacl was 170mM and 350mM respectively. The inhibitory activity of 170mM dissolution fraction was significantly higher than that of IL-1 and IL-2. The inhibitory activity of 350mM dissolution fraction was significantly higher than that of IL-1. The isoelectric point of the inhibitor of dissolution was 59, 52 ~ 53 at 170mM and 350mM, respectively. The inhibitory factor of 170mM dissolution fraction was decreased by acid treatment at pH 3.0, and decreased by heat treatment at 56℃ for 60 minutes. The inhibitory factor of 350mM dissolution fraction decreased by acid treatment at pH 3.0.
