1.新生児乾燥濾紙血アポB濃度測定による高脂血症マススクリ-ニングの実用化のため、乾燥濾紙血の保存条件を検討した。室温放置では2〜5日間で大幅な測定値の低下を認めた。が、原因はアポBの酸化、蛋白分解によることが明かとなった。グラディエント電気泳動法とWestern Blotting法により、LDLは室温放置により径の小さく比重の大きいLDLに変化、分解され、アポBの免疫抗原性も低下することを明らかにした。予め、酸化防止剤(ethylenediaminetetraacetic acis)、蛋白分解剤阻害剤(phenylmethylsulfonyl fluoride)で処理した乾燥濾紙を用い、血液採取後、密封遮光下で保存すると、2週間は充分に保存が可能であり、測定値の再現性も良好であった。2.これまで上記の処理濾紙を用い500検体の新生児乾燥濾紙血を入手し、アポB濃度を測定した。5名の高アポB血症(>40mg/dl)が同定できたが、再検後も高値であったものは3名(0.6%)であり、家族性高コレステロ-ル血症ヘテロ接合体を新生児期から同定できる可能性が示唆された。3.100人の新生児臍帯血のLDLを解析し、小型で高比重のLDLを有する新生児は、23名(23%)存在し、正常のLDLを有する新生児に比較して有意に家族に肥満を有するものが多く、またアポB/コレステロ-ル比も高かったことから、複合型高脂血症の早期診断の可能性が示唆された。
